With the SharePoint 2007 products, you must use the Alternate Access Mappings (AAM) to get SharePoint to "listen" for alternate host header values. Changing the host header in IIS will NOT fix the issue.

Make sure you check the DNS entries. I ran into that same problem and I fixed it looking at the DNS. Also, check what port your web app is on. If it's on a port other than 80, then you may need to enter in http://myMossServer:<portnumber>/sites/myPortal. If it's on a different port number...

It sounds like your Alternate Access Mappings (AAM) are not configured correctly or haven't been setup. AAMs will allow your web application to respond to different 'names' just as you described. I believe this TechNet article will give you some guidance: Configure alternate access mapping
